UPLC-MS/MS法测定新鲜烟草中的脱落酸和茉莉酸
Determination of Abscisic and Jasmonic Acids in Green Tobacco by UPLC-MS/MS

摘要: 为更加快速准确地测定烟草中的酸性植物激素,通过优化样品的前处理条件,建立了超高效液相色谱-串联质谱法(UPLC-MS/MS)测定烟草中脱落酸和茉莉酸的方法。结果表明:①脱落酸和茉莉酸的检出限分别为0.082和0.027 ng/mL,加标回收率均在96.4%~110.8%之间,脱落酸的日内和日间相对标准偏差(RSDs)均低于3.86%,茉莉酸的日内和日间RSDs均低于4.53%。②该方法样品用量仅为25 mg;所采用的负离子检测模式选择性高、干扰小。适用于烟草样品中酸性植物激素内源性水平的准确检测。Abstract: In order to determine acidic phytohormones in tobacco fast and accurately, an ultra-performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS) method for detecting abscisic acid(ABA) and jasmonic acid(JA) in tobacco was developed by improving the conditions of sample pretreatment. The results showed that: 1) The detection limits of ABA and JA were 0.082 and 0.027 ng/m L, respectively; their standard addition recoveries ranged from96.4% to 110.8%. The intra- and inter-day relative standard deviations(RSDs) of ABA were less than 3.86%, and those of JA were less than 4.53%. 2) Only 25 mg of tobacco sample were needed for detection; and the negative ion detection mode adopted by this method featured high selectivity and less interference. This method provides a basis for accurately detecting the endogenous levels of acidic phytohormones.
