烟草β-胡萝卜素羟化酶基因的转录表达模式及功能分析

Transcription Expression Pattern and Function Analysis of β-OHase Gene in Nicotiana tabacum

  • 摘要: 为了明确烟草β-胡萝卜素羟化酶(β-carotene hydroxylase,β-OHase)基因的转录表达模式和功能,进而调控烟草类胡萝卜素的代谢,通过Real-time PCR分析了普通烟草(Nicotiana tabacum)品种红花大金元盛花期的β-OHase基因在不同器官中的表达量。利用烟草脆裂病毒诱导的基因沉默(VIGS)体系抑制本氏烟草(Nicotiana benthamiana)中β-OHase基因的表达,在该基因成功沉默后,通过Real-time PCR检测其下游基因表达量,同时检测烟草中β-胡萝卜素、紫黄质和新黄质含量(质量分数)。结果显示,β-OHase基因在叶片中的表达量明显高于其他器官(P < 0.05)。与对照相比,在该基因沉默后,下游的紫黄质脱环氧化酶基因(VDE)和玉米黄质环氧化酶基因(ZE)的表达量明显降低,同时β-胡萝卜素含量显著升高,而紫黄质和新黄质含量则显著降低(P < 0.05)。

     

    Abstract: In order to clarify the transcription expression pattern and functions of β-carotene hydroxylase gene(β-OHase) in Nicotiana tabacum and further to regulate the metabolism of carotenoids in tobacco, the expression level of β-OHase in different tissues and organs of cv.Honghuadajinyuan(Nicotiana tabacum) at full flowering stage was studied by Real-time PCR.On account of the depression of β-OHase expression in Nicotiana benthamiana via the gene silencing system induced by tobacco rattle virus, the expression levels of downstream genes(such as VDE & ZE) were detected by Real-time PCR and the contents of carotenoids(β-carotene, violaxanthin and neoxanthin) in tobacco were determined after β-OHase silencing.The results showed that the expression level of β-OHase was significantly higher(P < 0.05) in leaves than in the other organs. Comparing with the control, the expression levels of VDE & ZE and the contents of violaxanthin and neoxanthin significantly decreased, while β-carotene content obviously increased(P < 0.05) after β-OHase silencing.
