烟支霉变微生物的鉴定及其生物学特性分析

  • 摘要: 为明确引起烟支霉变的微生物及其生物学特性,从霉变烟支中分离到4株霉菌。采用真菌形态学和rDNA-ITS分子标记相结合的方法,对这4株霉菌进行鉴定,并对其生物学特性进行研究。结果表明:① 引起烟支霉变的微生物主要为球孢枝孢、黄曲霉、桔青霉和米曲霉。② 4种霉菌对不同培养基、温度、湿度的适应性分别为黄曲霉 > 桔青霉 > 球孢枝孢 > 米曲霉、黄曲霉 > 球孢枝孢 > 桔青霉 > 米曲霉、黄曲霉 > 桔青霉 > 球孢枝孢 > 米曲霉。球孢枝孢在pH≥5.0时生长较好;黄曲霉在pH=4.0时生长最好;桔青霉适宜的pH范围较宽,在pH=3.0时也能很好地生长;米曲霉在pH=7.0时生长最好。4种霉菌的致死温度和处理时间分别为球孢枝孢75℃ 10 min,黄曲霉75℃ 10 min或80℃ 5 min,桔青霉75℃ 10 min,米曲霉65℃ 5 min。4种霉菌对碳源和氮源的利用情况均为黄曲霉 > 桔青霉 > 球孢枝孢 > 米曲霉。紫外线处理对烟支霉菌有一定的抑制作用,其抗紫外线的能力为球孢枝孢 > 黄曲霉 > 桔青霉 > 米曲霉。

     

    Abstract: To investigate the microorganisms resulted mildew cigarettes and their biological characteristics, four strains of mold isolated from mildew cigarettes were analyzed by observing the morphological characteristics of the mold and analyzing the sequences of rDNA-ITS, the biological characteristics of the mold were studied as well. The results showed that: 1) The main microorganisms from the mildew affected cigarettes were Cladosporium sphaerospermum, Aspergillus flavus, Penicillium citrinum and Aspergillus oryzae. 2) The four strains in the order of their adaptabilities to different culture mediums, temperatures and humidity were A. flavus > P. citrinum > C. sphaerospermum > A. oryzae, A. flavus > C. sphaerospermum > P. citrinum > A. oryzae and A. flavus > P. citrinum > C. sphaerospermum > A. oryzae, respectively. C. sphaerospermum grew well on culture medium with pH≥ 5; A. flavus grew best at pH of 4.0; P. citrinum had wide adaptability to pH and grew well at pH of 3; A. oryzae grew best at pH of 7. The lethal temperature of C. sphaerospermum, P. citrinum and A. oryzae was 75 ℃ for 10 min, 75 ℃ for 10 min and 65 ℃ for 5 min, respectively; that of A. flavus was 75 ℃ for 10 min or 80 ℃ for 5 min. The rank of the four strains for carbon and nitrogen source utilization were A. flavus > P. citrinum > C. sphaerospermum > A. oryzae. UV had a certain inhibitory effect on the microorganisms of the mold in mildew cigarettes, and the anti-ultraviolet abilities of the four strains were C. sphaerospermum > A. flavus > P. citrinum > A. oryzae.
