烟草青枯病拮抗菌的筛选及发酵条件试验
Screening and fermentation conditions of antagonistic bacterium against tobacco bacterial wilt

摘要: 为了筛选对烟草青枯病有拮抗作用的生防菌株,有效防治该病害,从青枯病发病较重田块的健康烟草根系周围土壤中取样,采用点接法筛选出6株对烟草青枯病有较强拮抗作用的菌株,其抑菌圈直径均>8 mm,其中菌株XC4抑菌活性较强且拮抗效果稳定。通过单因素试验进行了发酵条件的初步研究,结果表明,当发酵时间48 h,发酵温度30℃,pH值7,接种量2%,转速190 r/min,碳源为牛肉膏,氮源为胰蛋白胨时,菌株XC4发酵效果最好。发酵条件经初步优化,菌株XC4对青枯病菌的拮抗效果由原来的抑菌圈直径12.5 mm增至21.5 mm,比优化前提高了72%。Abstract: In order to screen antagonistic bacteria against tobacco bacterial wilt, soil samples were collected from the rhizosphere soil of seriously diseased tobacco plants, and six strains (XC1, XC2, XC3, XC4, XC5, DZ1) with strong antagonistic activity were identified by the point contact method. The diameters of antibacterial inhibition zone of the six strains were all above 8 mm. Among them the strain XC4 had a stronger antibacterial activity and stable antagonistic effect. The fermentation conditions were preliminarily studied by a single factor test. The results showed that the optimum fermentation conditions for XC4 were fermenting at 30℃, pH 7.0, inoculum concentration at 2%, 190 r/min for 48 hours with beef extract as carbon source and tryptone as nitrogen source. Under these conditions, the diameter of inhibition zone of XC4 against Ralstonia solanacearum increased by 72%, from 12.5 to 21.5 mm.
