基于高通量测序的废次烟末水提液中细菌群落分析

High-throughput bacterial analysis on aqueous extract of waste tobacco scrap

  • 摘要: 为了解造纸法再造烟叶生产过程中不同烟末提取液的微生物变化,利用Roche 454 FLX焦磷酸高通量测序法,比较废次烟末水提液中细菌16S rRNA基因序列的多样性,获得了废次烟末水提液中细菌种群的组成和丰度的变化。结果表明:在废次烟末水提液的原液(M1)、浓缩液(M2)及醇化液(M3)样品中,细菌种群组成非常丰富,其中占主导地位的细菌种群是厚壁菌门,而厚壁菌门中的优势菌属是乳杆菌属和赖氨酸芽孢杆菌属。从M1到M3的过程中,乳杆菌属丰度呈下降趋势,而赖氨酸芽孢杆菌属丰度呈上升趋势。基于高通量测序的细菌群落变化趋势可为改善造纸法再造烟叶的品质提供依据。

     

    Abstract: To investigate the variation of microorganisms in aqueous extract of waste tobacco scrap during paper-making process of reconstituted tobacco sheet materials, Roche 454 FLX pyrosequencing was adopted to analyze the diversity of 16S rRNA gene sequences of bacteria in the aqueous extract of waste tobacco scrap. The variation of bacterial communities and their abundances in the extracts was obtained. The results showed that:1) The aqueous extracts of waste tobacco scrap, including the original extract (M1), the concentrated extract (M2) and the aged extract (M3), harbored abundant levels of bacteria. The dominant phyla was Firmicutes. Lactobacillus and Lysinibacillus were the dominant genera of Firmicutes in all the samples. 2)It was also found that the abundance of Lactobacillus decreased from M1 to M3, while the abundance of Lysinibacillus increased. The variation of bacterial community based on high-throughout sequencing provides a scientific basis for improving the quality of paper-making reconstituted tobacco materials.
