UPLC-HRMS法测定无烟气烟草制品中的小分子醛

Analysis of small-molecule aldehydes in smokeless tobacco products by UPLC-HRMS

  • 摘要: 为评价无烟气烟草制品中小分子醛的含量状况,通过水-异己烷两相体系同步实现甲醛、乙醛、丙醛、丙烯醛、丁醛、巴豆醛及内标物的超声提取和2,4-二硝基苯肼(DNPH)衍生化,然后用Q-Exactive质谱仪检测分析物,建立了一种同时分析无烟气烟草制品中6种小分子醛的超高效液相色谱-高分辨质谱联用(UPLC-HRMS)方法。结果表明:①优化后的样品制备过程不超过30 min,样品分析时间仅为6min;②通过选择特征二级离子进行定量,排除了丙酮对丙醛、丁酮对丁醛的干扰;③各分析物的线性范围超过两个数量级,线性关系良好(R2>0.99),定量限为0.07~0.90 ng/mL,加标回收率在81.1%~107.6%之间,相对标准偏差(RSD)≤12.7%;④在无烟气烟草制品中检出的小分子醛主要是甲醛和乙醛。该方法简单、快速、可靠,适用于无烟气烟草制品中小分子醛的测定。

     

    Abstract: To investigate the contents of small-molecule aldehydes in smokeless tobacco products, such as formaldehyde, acetaldehyde, propionaldehyde, acrolein, butyraldehyde and crotonaldehyde, these targets and their internal standards were extracted under ultrasound and derived with 2, 4-dinitrophenylhydrazine(DNPH) simultaneously by a water-isohexane biphase system. The derivatives were then analyzed by Q-Exactive mass spectrometry with ultra-performance liquid chromatography-high resolution mass spectrometry(UPLC-HRMS) method. The result showed that:1)Preparation time was less than 30 min after optimization, and analysis time was reduced to 6 min by adopting C18 UPLC column. 2) The interferences of acetone and butanone to propionaldehyde and butyraldehyde were excluded by using characteristic secondary ion for quantitative analysis. 3)The linear ranges of analytes were over two orders of magnitude with good correlation coefficients (R2>0.99), the limits of quantification ranged from 0.07 to 0.90 ng/mL, and the recoveries were in the range of 81.1%-107.6% with relative standard deviations no more than 12.7%. 4)Formaldehyde and acetaldehyde were major small-molecule aldehydes in smokeless tobacco products. The established method is simple, rapid and reliable, it is suitable for the analysis of small-molecule aldehydes in smokeless tobacco products.
