Abstract:
In order to achieve high-throughput and accurate determination of starch in tobacco, a sample extraction vial was designed and used as a pre-treatment device to establish an ultra-fast method for determining starch. By using this extraction vial, the removal of interference components, the extraction and the filtration could be achieved without sample transfer. The results showed that:1) The sample extraction vial integrated the functions of sample extraction, filtration and waste removal, which significantly improved preparation speed. 2) The linear relationship of starch was good (
r=0.999 6). The mean recoveries ranged from 96.40% to 101.30%, and the intra-day and inter-day precision was less than 3.3%. 3) The contents of starch in eight gene edited tobacco samples were consistent with the data determined by the current tobacco industry standard method. The proposed method is simple, rapid, sensitive, reproducible and suitable for high-throughput starch detection in tobacco samples.
