RNAi vector construction and functional analysis of NtGGPPS4 in Nicotiana tabacum

  • 摘要: 为研究NtGGPPS4在合成烟草萜类化合物中的功能,构建了该基因的RNAi表达载体pHellsgate2-NtGGPPS4,通过农杆菌介导法进行遗传转化。使用荧光定量PCR技术检测转基因烟株中NtGGPPS4的表达量,采用溶剂萃取法从转基因烟株中提取西柏三烯醇和质体色素,并分别使用GC-MS、LC-MS测定其含量。结果表明,在NtGGPPS4表达量明显下降的转基因烟株中,α-西柏三烯二醇、β-西柏三烯二醇、新黄质、紫黄质、β-胡萝卜素、叶绿素a、叶绿素b和叶黄素的含量明显下降。表明NtGGPPS4参与调控了烟叶中西柏三烯醇和质体色素的生物合成。

     

    Abstract: To study the function of NtGGPPS4 in tobacco terpenoid synthesis, pHellsgate2-NtGGPPS4, an RNAi expression vector, was constructed and transferred by Agrobacterium-mediated transformation method. The transcript accumulation of NtGGPPS4 in transgenic tobacco plants was detected by qRT-PCR. α-and β-Cembrenediol and plastid pigments in transgenic tobacco plants were extracted by solvent extraction method. And the contents of α-and β-cembrenediol and plastid pigments were determined by GC-MS and LC-MS, respectively. The results showed that the expression level of NtGGPPS4 significantly decreased in transgenic tobacco plants. Comparing with the control, the contents of α-and β-cembrenediol and plastid pigments (neoxanthin, violaxanthin, β-carotene, chlorophyll a, chlorophyll b and lutein) significantly decreased. The results suggested that NtGGPPS4 was involved in the regulation of cembrenediol and plastid pigment biosynthesis in Nicotiana tabacum.
