超高效液相色谱法测定烟草中顺-冷杉醇

Determination of cis-abienol in tobacco with ultra-high performance liquid chromatography

  • 摘要: 为准确测定烟草样品顺-冷杉醇的含量(质量分数),通过优化提取方法及液相色谱条件,建立了烟草粉末样品中顺-冷杉醇提取及应用超高效液相色谱-紫外(UPLC-UV)检测的方法,并采用该方法对特香型烤烟品种大白筋599烘烤前后的15份烟叶样品进行了测定。结果表明:①顺-冷杉醇含量在1.60~80.00 μg/mL范围内,工作曲线线性良好(r2=0.999 9);②加标回收率为97.63%~109.80%,相对标准偏差(RSD)为2.05%~3.69%,方法检出限为4.00 μg/g,定量限为13.30 μg/g,RSD为3.43%。③烟草中的顺-冷杉醇在调制过程中大部分降解,降解率为27.92%~89.23%,平均为63.66%。该方法样品前处理简单,分离度好,精密度和准确性较高,适用于烟草中顺-冷杉醇的批量检测。

     

    Abstract: In order to accurately determine the content of cis-abienol in tobacco samples, an ultra-high performance liquid chromatography-ultraviolet (UPLC-UV) method was established by optimizing the extraction method and liquid chromatography conditions. The established method was used to determine the cis-abienol in 15 samples of flue-cured tobacco cv. Dabaijin599 with special flavor type before and after curing. The results showed that:1) The linear relationship of the curve was good (r2=0.999 9) in the range of 1.60-80.00 μg/mL of cis-abienol. 2) The spiked recoveries of samples were from 97.63% to 109.80%, the relative standard deviations (RSDs) ranged from 2.05% to 3.69%, the limit of detection was 4.00 μg/g and the limit of quantitation was 13.30 μg/g with the reproducibility RSD of 3.43%. 3) During the process of tobacco curing, cis-abienol in tobacco mostly degraded at a rate of 27.92%-89.23%, averaged at 63.66%. This method features a simple sample pretreatment, good separation, a high precision and accuracy, and thus suitable for the detection of cis-abienol in tobacco.
