Analysis of characteristic aroma components in tobacco aroma mutant by HS-SPME arrow-GC/MS combined with chemometrics

  • 摘要: 为明确特征香韵烟草突变体(萨姆逊香)与对照中烟100中香气成分的差异,优化烟草挥发性香气成分检测方法,通过考察箭型固相微萃取(SPME arrow)萃取头类型、平衡时间、萃取时间和萃取温度等样品前处理条件,建立了测定烟草中挥发性香气成分的顶空-箭型固相微萃取-气相色谱质谱联用(HS-SPME arrow-GC/MS)方法;采用该方法对两个材料进行了检测,并结合主成分分析(PCA)和偏最小二乘-判别分析(PLS-DA)方法分析了萨姆逊香烟草突变体中的特征香气成分。结果表明:① SPME arrow优化萃取条件为120 μm×20 mm活性炭/聚二甲基硅氧烷(CAR/PDMS)萃取头,70℃下平衡40 min,萃取40 min;在此条件下,质控样(QC)中相对标准偏差 < 30%的色谱峰数量占总峰数量的89.3%。②从两个材料中共筛选出差异香气成分33种,萨姆逊香突变体材料中29种成分的含量高于中烟100。该方法操作简单,重现性好,结果稳定可靠,适用于烟叶特征香气成分的分析。

     

    Abstract: In order to clarify the differences of aroma components between the characteristic note tobacco mutant (Samson) and Zhongyan 100 (ZY100, the control) and to optimize the detection of volatile aroma components in tobacco, a headspace-solid phase microextraction arrow and gas chromatography-mass spectrometry (HS-SPME arrow-GC/MS) method was established by investigating the type of SPME arrow, equilibrium time, extraction time and extraction temperature. The aroma components in two tobacco materials were analyzed by this method, and the characteristic aroma components in Samson tobacco mutant were analyzed by principal component analysis (PCA) and partial least squares-discrimination analysis (PLS-DA). The results showed that:1) The optimized extraction conditions of SPME arrow were 120 μm×20 mm CAR/PDMS, equilibrium for 40 min and extraction for 40 min at 70℃. Under the conditions, the number of peaks with a relative standard deviation of less than 30% in the quality control (QC) samples accounted for 89.3% of the total peaks. 2) A total of 33 aroma components were detected in the two materials, among them the contents of 29 components in Samson tobacco mutant were higher than those in ZY100. This method has the advantages of easy operation, good reproducibility, stability and reliability, it is suitable for analyzing the characteristic aroma components in tobacco leaves.
