分泌型烟草腺毛形态和叶面化学成分的比较

Comparative studies on secretory glandular trichome morphology and leaf surface chemistry between tobacco varieties

  • 摘要: 西柏烷二萜和蔗糖酯是腺毛分泌物的主要成分。为了明确不同类型腺毛的分泌物差异,以不同腺毛类型的烟草品系TI1068和TI35为材料,分析了表皮毛的形态结构、叶面化学成分、西柏烷二萜合成基因的表达水平以及蚜虫的嗜好性。表皮毛形态学观察发现,TI1068和TI35同时具有长柄腺毛、短柄腺毛和保护毛3种表皮毛类型;TI1068以长柄分泌型腺毛为主,含有微量保护毛;TI35以保护毛为主,含有微量长柄腺毛;TI1068和TI35的短柄腺毛数量无显著差异。叶面化学成分检测结果表明,TI1068和TI35均含有西柏烷二萜、蔗糖酯和烷烃;TI1068叶面中3类物质总量是TI35的10倍;TI1068的叶面主要化学成分是西柏烷二萜和蔗糖酯,而TI35的叶面主要成分是烷烃,TI1068和TI35中烷烃的种类和总量差异不明显。西柏烷二萜合成基因GGPPSCBTSCYP71D16在TI1068中的表达水平均高于TI35。蚜虫嗜好性选择结果显示,蚜虫对TI1068的嗜好性极显著低于TI35。

     

    Abstract: Cembranoids and sucrose esters are the main secretions of glandular trichomes. In order to explore the characteristics of chemical components in different tobacco glandular trichome secretions, tobacco varieties TI1068 and TI35 (Nicotiana tabacum), which have different types of glandular trichomes, were used to analyze the morphological structure of trichomes, the chemical components on leaf surface, the expression level of genes related to cembranoid biosynthesis and aphid preference. Trichome morphological observations showed that TI1068 and TI35 had three types of trichomes:long stalk glandular trichomes, short stalk glandular trichomes, and protective trichomes. TI1068 mainly had long stalk glandular trichomes and few protective trichomes, while TI35 mainly had protective trichomes and few long stalk glandular trichomes. There was no significant difference in the number of short stalk glandular trichomes between TI1068 and TI35. Leaf surface chemical analysis indicated that both TI1068 and TI35 contained cembranoids, sucrose esters and alkanes, while the total content of the three chemicals in TI1068 was 10 times as that of TI35. The main components on the leaf surface of TI1068 were cembranoids and sucrose esters, while those of TI35 were alkanes. There was no obvious difference in the types and total amount of alkanes between TI1068 and TI35. The expression levels of GGPPS, CBTS and CYP71D16 related to cembranoid biosynthetic were higher in TI1068 than those in TI35. Aphid preference test showed that aphids significantly preferred TI35 rather than TI1068.
