Rapid screening of nicotine converters from tobacco seedlings

  • 摘要: 为快速鉴别烟草群体中的烟碱转化株,开发了一种基于气相色谱-质谱的烟草幼苗烟碱转化株筛选方法。该方法提供了非孵化和孵化两种策略。采用非孵化策略时,从采样到分析完成仅需5 ~ 6 min,该策略可在损失9%非转化株幼苗的前提下完全去除转化株;采用孵化策略时,只需将非孵化策略中所采样品在37 ℃下封闭培养12 h后进行提取和分析。孵化策略实现了本实验所涉及的幼苗群体(共221株)中转化株和非转化株的完全分离。本研究中所建立的筛选方法方便、高效,可为批量快速识别幼苗群体中烟碱转化株提供技术保障。

     

    Abstract: In order to rapidly identify nicotine converters in tobacco plants, a screening method based on gas chromatography-mass spectrometry was developed, and two strategies (with and without incubation) were proposed. The non-incubation strategy took about 5-6 min from sampling to analysis. With this strategy, the complete removal of converters could be achieved with the sacrifice of 9% non-converters. For the incubation strategy, in-tube samples were cured at 37 ℃ for 12 h before the extraction and analysis. The complete separation of converters from non-converters in 221 seedlings was achieved by using the incubation strategy. The developed method provides a technical support for the fast and efficient identification of nicotine converters from tobacco seedlings.
