Abstract:
In order to establish a rapid and quantitative test method to measure carbendazim residues in tobacco leaves for field detection, carbendazim test strip based on colloidal gold immunochromatography technology and colloidal gold reader were developed and evaluated. The influence of temperature on the quantitative results by the colloidal gold test strip was clarified, and the standard curve of the colloidal gold reader was set by controlling the detection temperature with a constant temperature incubator. The results showed that: 1) The detection limit of the developed quantitative colloidal gold test strip was 0.017 mg/kg with the detection range of 0.020-2.0 mg/kg and the quantitative range of 0.020-0.50 mg/kg. The standard recovery was 95.6%-128.9% and intra assay coefficient of variation was 6.2%-11.2%. 2) Compared with the results tested by using high performance liquid chromatography tandem mass spectrometry, the test result accuracy of negative samples was higher than 95%, and the test results of positive samples had a good linear correlation in the range of 0.020-0.50 mg/kg. 3) The test strip had no cross reaction with similarly structured drugs, therefore it could be used to detect the carbendazim residues in tobacco leaves. The developed quantitative test strip had the advantages of simple to use, fast detection speed, high accuracy and good stability, and is thus suitable for the detection of large quantities of samples in primary laboratories and field detection.
