一株枯草芽胞杆菌L249的分离鉴定及对烟草赤星病的盆栽防效

Isolation and identification of Bacillus subtilis strain L249 and its control effect on tobacco brown spot disease in pot experiments

  • 摘要: 为筛选出对烟草赤星病具有生防潜力的拮抗细菌,以链格孢菌(Alteraria alternata)为靶标菌,采用5点取样法在赤星病发生严重地块采集健康烟株根际土壤,通过梯度稀释分离法和平板对峙法筛选得到一株具有生防作用的拮抗菌L249,对链格孢菌抑菌率达72.27%,经菌落形态、生理生化和分子生物学鉴定为枯草芽胞杆菌(Bacillus subtilis)。该菌能产生蛋白酶、纤维素酶和硝酸还原酶等生防相关酶类。利用比浊法测定菌株L249的生长速率,结果表明该菌培养4 h后进入对数生长期,14 h后进入稳定生长期。盆栽试验结果表明,菌株L249发酵液能有效抑制烟草赤星病的发生,相对防效达60.66%,与10%苯醚甲环唑水分散粒剂的相对防效差异不显著。枯草芽胞杆菌L249可有效防治烟草赤星病,具有较好的生防潜力。

     

    Abstract: In order to screen the antagonistic bacteria strains with biocontrol effects against tobacco brown spot disease, taking Alteraria alternata as the target bacteria, the soil samples were collected by a 5-point sampling method from the rhizospheric soil of healthy tobacco plants in fields with serious tobacco brown spot disease. A bacterium strain, L249, against Alteraria alternata with an inhibition rate of 72.27% was screened by the gradient dilution isolation method and plate confrontation method. The strain L249 was identified as Bacillus subtilis through colony morphological, physiological, biochemical, and molecular biological tests and could produce biocontrol-related enzymes such as protease, cellulose, and nitrate reductase. The growth rate of strain L249 was measured by turbidimetry, and the results showed that the bacteria entered the logarithmic growth phase after 4 h and entered the stable growth phase after 14 h. The results of pot experiments revealed that fermentation broth of strain L249 could effectively inhibit the occurrence of tobacco brown spot disease with a relative control efficiency of 60.66%, which was not significantly different from that of 10% difenoconazole WDG. Thus, Bacillus subtilis strain L249 can effectively control tobacco brown spot disease and has a good potential for biological control.
