雪茄烟中霉菌计数方法的适用性评价

摘要: 为明确霉菌计数方法对雪茄烟的适用性, 采用孟加拉红琼脂培养基、改良马铃薯葡萄糖琼脂培养基和SYMPHONY Agar快速显色培养基对13种雪茄烟进行霉菌计数, 评价3种培养基在不同接种方式条件下雪茄烟中霉菌计数结果的一致性, 并对各霉菌计数方法进行比较。结果表明:①同种接种方式条件下, 3种培养基的霉菌计数结果的一致性较好;②相同培养基条件下, 平板涂布法操作更简便, 菌落分布均匀, 典型菌落更易判读;③3种霉菌计数培养基相比, SYMPHONY Agar快速显色培养基培养周期短, 霉菌典型菌落大小均匀, 更容易计数。因此, SYMPHONY Agar快速显色培养基和平板涂布法接种方式更适合于雪茄烟中的霉菌计数。Abstract: To investigate the applicability of mould counting methods for cigar, the degrees of mould in 13 cigar samples were counted by using rose bengal agar medium, modified potato dextrose agar medium, and SYMPHONY Agar rapid chromogenic medium. The consistency of the mould counting results on the three media with different inoculation methods was evaluated, and the mould counting methods were compared. The results showed that: 1)The consistency of the mould counting results on the three media was better when adopting the same inoculation method. 2)On the same medium, spread plate method was more convenient in operation, the colonies distributed evenly, and the typical colonies were easier to be interpreted. 3) Comparing with the other two media, SYMPHONY Agar rapid chromogenic medium required a shorter culture period, the sizes of typical mould colonies were uniform, and the mould counting was easier. Therefore, SYMPHONY Agar rapid chromogenic medium combined with the spread plate method is more suitable to the mould counting of cigar.
