不同采烤方式上部烟叶外观质量及抗氧化特性分析

Appearance quality and antioxidant properties of upper tobacco leaves harvested and cured by different methods

  • 摘要: 为明确带茎采烤上部烟叶的品质特点及其形成机理,以云烟87为试材分析了上部烟叶带茎采烤和逐叶采烤烟叶外观质量特点、烘烤阶段干球温度关键稳温点的烟叶显微结构和抗氧化能力的差异。结果表明:①带茎采烤上部烟叶外观质量显著好于逐叶采烤,其颜色特征黄色面积、L*值、b*值较高而a*值较低。②随着烘烤阶段的推进,在关键稳温点,带茎采烤上部烟叶组织细胞消融皱缩破坏程度滞后于逐叶采烤。③随着烘烤阶段的推进,烟叶细胞膜脂质过氧化物(LPO)、丙二醛(MDA)含量(质量分数)总体均呈现升高趋势,均在定色阶段46 ℃末达到最高,且带茎采烤上部烟叶比逐叶采烤分别降低21.02%和32.01%;烟叶过氧化氢(H2O2)含量和超氧阴离子(O2-·)产生速率总体均呈现逐渐升高的趋势,且带茎采烤上部烟叶在变黄阶段均低于逐叶采烤,在定色阶段46 ℃末则显著提高。④随着烘烤阶段的推进,带茎采烤上部烟叶超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性总体高于逐叶采烤烟叶;带茎采烤上部烟叶谷胱甘肽还原酶(GR)活性呈逐步升高又降低的趋势,在变黄阶段42 ℃末达到最高,而逐叶采烤的烟叶GR活性呈慢-快-慢升高的趋势,在定色阶段46 ℃末达到最高,且变黄阶段带茎采烤上部烟叶GR活性高于逐叶采烤烟叶;带茎采烤上部烟叶总抗氧化能力(T-AOC)总体呈逐步升高的趋势,且高于逐叶采烤烟叶,在变黄阶段40 ℃末、定色阶段46 ℃末,带茎采烤与逐叶采烤上部烟叶T-AOC的差异达到显著水平。总之,带茎采烤上部烟叶的外观质量较好,这与烘烤过程中烟叶组织细胞消融破坏程度滞后和抗氧化能力较强密切相关。

     

    Abstract: In order to clarify the quality and their formation mechanism of the upper leaves harvested with stalks, Yunyan 87 was used as a test material to analyze the quality characteristics of the upper leaves harvested and cured with stalks and the upper leaves harvested and cured leaf by leaf and the differences in the microstructure, antioxidant capacity and dry bulb temperature of the leaves at the key temperature points during the curing stage. The results showed that: 1) Compared to the leaves without stalks, the appearance quality of the leaves with stalks was significantly better, the area with characteristic yellow color was larger, the L* and b* values were higher, and the a* value was lower. 2) As the curing process progressed, the tissue and cell morphology of the upper leaves with stalks were destroyed by ablation, and the degree of shrinkage lagged behind that of the upper leaves harvested without stalks at various critical temperature points. 3) As the curing process progressed, the mass fractions of lipid peroxide (LPO) and malondialdehyde (MDA) in the leaves generally increased and peaked at the end of the 46 ℃ interval in the color fixing stage, and those in leaves with stalks were 21.02% and 32.01% lower than those in leaves without stalks. The mass fraction of hydrogen peroxide (H2O2) and the production rate of superoxide anion (O2-·) in leaves increased gradually, which were lower in the yellowing stage and higher at the end of the 46 ℃ interval in the color fixing stage in leaves with stalks than in leaves without stalks. 4) As the curing process progressed, the activities of superoxide dismutase (SOD) and catalase (CAT) in leaves with stalks were higher than those in leaves without stalks. The activity of glutathione reductase (GR) in leaves with stalks first increased gradually, then decreased and peaked at the end of the 42 ℃ interval in the yellowing stage; while that of leaves with stalks slowly and gradually increased, and then rapidly and slowly, before peaked at the end of the 46 ℃ interval in the color fixing stage, and the GR activity of leaves with stalks was higher than that of leaves without stalks in the yellowing stage. The total antioxidant capacity (T-AOC) of tobacco leaves generally showed a gradual upward trend, that of leaves with stalks was higher than that of leaves without stalks, and the difference reached a significant level at the end of the 40 ℃ interval in the yellowing stage and at the end of the 46 ℃ interval in the color fixing stage. In conclusion, the appearance quality of the upper leaves harvested and cured with stalks was better, which was closely related to the delayed destruction of the tissue cells and the strong antioxidant capacity of the leaves during the curing process.
