烟草多主棒孢霉中携带真菌病毒的种类鉴定及特征分析

Speciation and characterization of mycoviruses in Corynespora cassiicola on tobacco

  • 摘要: 为发掘鉴定多主棒孢霉中的真菌病毒资源,利用高通量测序和宏病毒组分析技术,对来源于烟叶的11株多主棒孢霉菌株进行真菌病毒的发掘和鉴定。基于获得的病毒序列信息进行结构蛋白预测和系统发育分析,并明确了不同病毒在试验菌株中的侵染分布情况。结果表明:①菌株中共携带有5种真菌病毒,包括3种双链RNA(dsRNA)病毒、1种正义单链RNA(+ssRNA)病毒和1种负义单链RNA(-ssRNA)病毒。②蛋白比对和系统发育分析表明,这5种病毒中有2种属于整形病毒科(Totiviridae),其余3种分别属于双分体病毒科(Partitiviridae)、裸露RNA病毒科(Narnaviridae)和真菌蛇形病毒科(Aspiviridae)。③RT-PCR检测结果表明,这些菌株的带毒率为64%,并且2个菌株存在病毒的复合侵染。

     

    Abstract: To discover and identify the mycovirus sources in Corynespora cassiicola strains, high-throughput sequencing and mycovirome analysis were used to discover and identify the mycoviruses in eleven C. cassiicola strains isolated from tobacco leaves in China. Based on the obtained viral sequence data, structural protein prediction and phylogenetic analyses were performed, and the infections of different viruses in the test strains were clarified. The results showed that: 1) These strains carried a total of five mycoviruses, including three double-stranded RNA (dsRNA) viruses, one positive-sense single-stranded RNA (+ssRNA) virus and one negative-sense single-stranded RNA (-ssRNA) virus. 2) Coding protein alignments and phylogenetic analyses indicated that two of these five viruses belonged to Totiviridae, while the other three belonged to Partitiviridae, Narnaviridae, and Aspiviridae, respectively. 3) RT-PCR detection results showed that the virus carrying rate of these strains was 64%, and two strains had mixed infection of the viruses.
