烟草转录因子NtNAC029基因的克隆及表达模式分析

Cloning and expression pattern analysis of transcription factor NtNAC029 gene from Nicotiana tabacum

  • 摘要: 为明确NtNAC029基因在调控烟草叶片衰老和类黄酮合成过程中的生物学功能,根据拟南芥同源基因信息,从栽培烟草K326中克隆获得1个NAC转录因子基因(NtNAC029),该基因编码区及编码氨基酸具有NAC家族保守的序列结构和蛋白功能域。NtNAC029基因表达具有明显的组织和时期特异性,其在烟草叶片中的表达量随叶片成熟衰老逐渐升高,至叶片衰老后期达到最大值,而在烟草蒴果中的表达量则在发育前期急剧升高,之后逐渐降低,表明该基因可能参与调控烟草叶片衰老和果实发育等生理过程。NtNAC029基因表达对光照较为敏感,在光照培养条件下该基因表达量显著降低,黑暗处理该基因表达量则显著升高。此外,盐、脱落酸(ABA)和茉莉酸甲酯(MeJA)等处理均能显著诱导NtNAC029基因的表达,这表明NtNAC029可能在响应介导多种环境因子调控烟草叶片衰老过程中发挥着重要作用。

     

    Abstract: To elucidate the biological function of NtNAC029 gene in the regulation of leaf senescence and flavonoid biosynthesis in tobacco, a NAC transcription factor gene (NtNAC029) was cloned from cultivated tobacco K326 based on information from the homologous gene in Arabidopsis thaliana. The coding region and the encoded amino acids of this gene showed conserved sequence structures and protein domains of the NAC family. The expression of the NtNAC029 gene showed significant tissue and stage specificities. Its expression level in leaves gradually increased with leaf maturity and senescence, and reached its maximum value at the late stage of leaf senescence. However, its expression level in capsules increased sharply at the early stage of development and then gradually decreased, indicating that this gene may be involved in regulating important physiological processes such as tobacco leaf senescence and fruit development. The expression of NtNAC029 gene was more sensitive to light, and the gene expression level was significantly decreased under light culture conditions, while darkness treatment significantly upregulated the gene expression level. In addition, salt, abscisic acid (ABA) and methyl jasmonic acid (MeJA) could significantly induce the expression of NtNAC029 gene, indicating that NtNAC029 may play an important role in the response to multiple environmental factors in the regulation of tobacco leaf senescence.
