A novel glucoarabinogalactan from tobacco leaves: Extraction, purification and structural characterization

  • 摘要: 为了促进烟草资源的高附加值转化利用,采用热水浸提法从低品质烟叶中提取烟叶多糖,经离子交换层析和凝胶柱层析分离纯化得到一种新型葡阿拉伯半乳聚糖(WTLP2b),采用离子色谱、凝胶过滤色谱、气相色谱-质谱联用、核磁共振波谱等分析技术对WTLP2b的结构进行了详细解析。结果表明:①WTLP2b由半乳糖(Gal,55%)、阿拉伯糖(Ara,33%)和葡萄糖(Glc,12%)组成,分子量为62.0 kDa,采用甲基化衍生和核磁共振波谱解析发现WTLP2b的主链骨架由→6)-β-D-Galp-(1构成,而侧链由β-D-Galp-(1→4)-β-D-Galp-(1→、α-L-Araf-(1→4)-β-D-Glcp-(1→和α-L-Araf-(1→5)-α-L-Araf-(1→组成,通过→6)-β-D-Galp-(1→的O-3键连接于主链。②WTLP2b以片状结构为主,且呈现聚集状态。③WTLP2b的热稳定性相对较好。本研究结果表明烟叶可以作为葡阿拉伯半乳聚糖的天然来源。

     

    Abstract: To promote value-added transformation and utilization of tobacco resources, a hot water extraction method was used to extract polysaccharides from low-quality tobacco leaves. A novel glucoarabinogalactan, named WTLP2b, was isolated and purified by ion exchange chromatography and gel column chromatography. The structure of the WTLP2b sample was characterized using various analytical techniques, including ion chromatography, gel filtration chromatography, gas chromatography- mass spectrometry and nuclear magnetic resonance spectroscopy. The results showed that: 1) The WTLP2b obtained consisted of galactose (55%), arabinose (33%) and glucose (12%), with a molecular weight of 62.0 kDa. Methylation derivation and NMR analysis revealed that WTLP2b's main chain was composed of →6)-β-D-Galp-(1→, while the side chains consisted of β-D-Galp-(1→4)-β-D-Galp-(1→, α-L-Araf-(1→4)-β-D-Glcp-(1→, and α-L-Araf-(1→5)-α-L-Araf-(1→, connected to the main chain via O-3 bond of →6)-β-D-Galp-(1→. 2) The WTLP2b sample mainly showed a sheet-like structure with aggregated morphology. 3) The thermal stability of the WTLP2b sample was relatively good. The results of this study show that tobacco leaves can serve as a natural source of glucoarabinogalactan.
