3株生防菌对烟草丝囊霉根腐病病原菌的抑制作用及防治效果

Inhibitory and control effects of three biocontrol bacterial strains on tobacco root rot caused by Aphanomyces iridis

  • 摘要: 为筛选烟草丝囊霉根腐病的高效生防菌株,采用平板对峙培养法测定贝莱斯芽孢杆菌(Bacillus velezensis)Ba-0321、多粘类芽孢杆菌(Paenibacillus polymyxa)LB-9和棘孢木霉(Trichoderma asperellum)Tr-0111对烟草丝囊霉根腐病病原菌鸢尾丝囊霉菌(Aphanomyces iridis)的拮抗效果,通过菌丝生长速率法测定3株生防菌菌液、发酵滤液及挥发性代谢物对鸢尾丝囊霉菌菌丝生长的抑制作用,并通过室内漂浮育苗接种试验分析生防菌发酵液对烟草丝囊霉根腐病的防治效果。结果表明,3株生防菌对鸢尾丝囊霉菌均有拮抗效果。贝莱斯芽孢杆菌Ba-0321、多粘类芽孢杆菌LB-9和棘孢木霉Tr-0111菌液浓度分别为1 × 107、1 × 108和1 × 106 cfu/mL时,对鸢尾丝囊霉菌的菌丝生长抑制率分别为86.70%、85.32%和92.63%。其中,棘孢木霉Tr-0111对鸢尾丝囊霉菌菌丝生长的室内毒力最强,EC50值为3.59 × 102 cfu/mL,其次是贝莱斯芽孢杆菌Ba-0321和多粘类芽孢杆菌LB-9,EC50值分别为1.74 × 104和1.41 × 105 cfu/mL。3株生防菌的发酵滤液和挥发性代谢物对鸢尾丝囊霉菌的菌丝生长均有抑制作用。其中,贝莱斯芽孢杆菌Ba-0321发酵滤液和挥发性代谢物对鸢尾丝囊霉菌的菌丝生长抑制率最高,分别为42.73%和16.31%。室内漂浮苗接种试验结果表明,棘孢木霉Tr-0111发酵液处理的烟草丝囊霉根腐病相对防效为66.53%,显著(P < 0.5)高于贝莱斯芽孢杆菌Ba-0321(51.69%)和多粘类芽孢杆菌LB-9(40.95%)。

     

    Abstract: To screen and identify efficient biocontrol bacterial strains against tobacco root rot caused by Aphanomyces iridis, the antagonistic effects of Bacillus velezensis Ba-0321, Paenibacillus polymyxa LB-9 and Trichoderma asperellum Tr-0111 were determined by plate confrontation cultivation. The inhibitory effects of bacterial solutions, fermentation broths and volatile metabolites of the three biocontrol strains on mycelial growth of A. iridis were determined using mycelial growth rate methods. The control effects of the fermentation broths of these strains on tobacco root rot caused by A. iridis were analyzed by inoculation experiments using floating seedlings. The results showed that all three biocontrol strains had antagonistic effects on A. iridis. When the solution concentrations of B. velezensis Ba-0321, P. polymyxa LB-9 and T. asperellum Tr-0111 were 1 × 107, 1 × 108 and 1 × 106 cfu/mL, the inhibition rates on mycelial growth of A. iridis were 86.70%, 85.32% and 92.63%, respectively. Among the tested strains, T. asperellum Tr-0111 had the strongest inhibitory effect against A. iridis with an EC50 of 3.59 × 102 cfu/mL. The EC50 values of B. velezensis Ba-0321 and P. polymyxa LB-9 were 1.74 × 104 and 1.41 × 105 cfu/mL, respectively. The fermentation broths and volatile metabolites of the three biocontrol strains also had inhibitory effects on the mycelial growth of A. iridis, with those of B. velezensis Ba-0321 showing the highest inhibition rates of 42.73% and 16.31%, respectively. In the floating seedling inoculation experiment, the relative control effect of the fermentation broth of T. asperellum Tr-0111 on tobacco root rot caused by A. iridis was 66.53%, significantly (P < 0.5) higher than that of B. velezensis Ba-0321 (51.69%) and P. polymyxa LB-9 (40.95%).
