烟用料液贮存中微生物群落演替及山梨酸钾对致腐菌的抑制效果

Microbial community succession during the storage of tobacco casing and the inhibitory effect of potassium sorbate against spoilage microorganisms

  • 摘要: 为探究烟用料液贮存过程中的微生物群落演替过程以及山梨酸钾对优势致腐菌的抑制效果,采用宏基因组学与分子生物学技术,系统分析了烟用料液贮存过程中微生物群落的动态变化,鉴定了优势致腐菌并阐明其生物学特性,考察了山梨酸钾对该菌生长的抑制作用。结果表明:在贮存第7天,烟用料液菌落总数和酵母菌数相较于第5天分别增加了4.7×103和2.8×103 CFU/mL,并伴随出现酸香、产气及黄色菌膜等现象。在贮存0~5 d内,烟用料液微生物物种丰富度和多样性呈上升趋势;但至第7天,二者均显著下降,群落均匀度降低,其结构转为由少数物种主导,其中威克汉姆酵母属(Wickerhamomyces)逐渐成为优势菌群。经鉴定,该属中的主要致腐菌为异常威克汉姆酵母(Wickerhamomyces anomalus)。该菌株的最适生长条件为30 ℃、pH值6.0,最佳碳源为蔗糖、果糖和葡萄糖。抑菌结果显示,山梨酸钾对异常威克汉姆酵母的抑制作用呈现浓度依赖性,在1.00 mg/mL浓度下,菌株生长出现明显延迟,随着山梨酸钾浓度梯度升高,菌株生长逐渐停滞,其最小抑菌浓度(MIC)确定为8.00 mg/mL。

     

    Abstract: To clarify microbial community succession and the inhibitory effect of potassium sorbate on the dominant spoilage microorganisms of tobacco casing during storage, metagenomics and molecular biological technologies were employed to systematically investigate the dynamic changes in microbial community, identify the dominant spoilage microorganism, elucidate its biological characteristics, and further evaluate the inhibitory effects of potassium sorbate against the strain. The results showed that on the 7th day of storage, the total viable count and yeast count in the tobacco casing increased by 4.7×103 and 2.8×103 CFU/mL, respectively, compared to those on the 5th day, accompanied by the emergence of sour odor, gas production and the formation of a yellow microbial film. Within 0-5 days of storage, the richness and diversity of microbial species in tobacco casing showed an upward trend but a significantly decrease on day 7, with reduced community uniformity. The microbial community structure was dominated by a limited number of species, with Wickerhammyces gradually becoming the dominant bacterial group. The dominant spoilage microorganism was identified as Wickerhamomyces anomalus. The optimal growth conditions for this strain were 30 ℃ with a pH value of 6.0, and the best carbon sources were sucrose, fructose, and glucose. The antibacterial results revealed that the inhibitory effect of potassium sorbate on W. anomalus exhibited concentration dependence. At a concentration of 1.00 mg/mL, a significant delay in the growth of the strain was observed. As the concentration gradient of potassium sorbate increased, the growth of the strain gradually stagnated, and the minimum inhibitory concentration (MIC) was determined to be 8.00 mg/mL.
