产烟碱脱氢酶菌株节杆菌Z3的发酵条件研究

Studies on Fermentation Conditions for Nicotine Dehydrogenase Producing Arthrobacter Z3

  • 摘要: 以卷烟厂堆积烟尘的土壤中分离得到的产烟碱脱氢酶菌株节杆菌Z3(Arthrobacter Z3)为初发菌株,通过正交试验,对其发酵培养基进行优化,并对发酵条件(初始pH值、接种量、温度、转速、最佳发酵时间)进行了研究。获得的最佳摇瓶发酵配方为:2.5%烟末提取液、0.1%豆粉、0.1%硫酸铵、1.33%K2HPO4·3H2O、0.4%KH2PO4、0.02%MgSO4·7H2O、0.004%MnSO4·4H2O、0.002%CaCl2·2H2O。最适发酵产酶条件为:温度30℃,摇床转速220 r/min,发酵时间9h,初始pH7.0,接种量6%。

     

    Abstract: Arthrobacter Z3,which produces nicotine dehydrogenase,was isolated from the soil in a disposal site of tobacco dust in the cigarette factory.The culture medium of Z3 was optimized with orthogonal experiment,and its fermentation conditions,including initial pH value,inoculum amount,temperature,rotation speed and fermentation time,were tested.According to orthogonal experiment,the optimum medium composition was 2.5% tobacco dust extract,0.1% soybean powder,0.1%(NH4)2SO4,1.33% K2HPO4·3H2O,0.4% KH2PO4,0.02% MgSO4·7H2O,0.004% MnSO4·4H2O,and 0.002% CaCl2·2H2O,and the optimum fermentation conditions were:fermenting for 9 h at 30℃,rotation speed of the shaker 220 r/min,initial pH 7.0,and inoculum amount 6%.
