高浓度烟碱降解菌的筛选、鉴定及降解特性

Screening,Identification and Degradation Characteristics of High Concentration Nicotine Degradation Bacterium Strain

  • 摘要: 以烟碱为唯一碳氮源分别从武汉卷烟厂废弃物和山东淄博地区多年植烟的土壤中分离出152株降解烟碱的细菌分离株,进一步筛选得到高浓度烟碱降解菌ZB-16A。经常规形态学、生理生化特征、16SrDNA序列分析和系统发育树研究表明,ZB-16A分离株属于恶臭假单胞菌;ZB-16A在温度30℃,培养基初始pH7.0,摇床转速为150rpm时,可耐受6000mg/L的烟碱,在48h内可完全降解烟碱;当烟碱初始浓度为7000mg/L时,该降解菌生长受到抑制。

     

    Abstract: Utilizing nicotine as the sole source of carbon and nitrogen,152 bacterial strains were isolated from tobacco growing soils in Zibo,Shandong Province and tobacco waste from Wuhan Cigarette Factory,respectively,and a high concentration nicotine degradation bacterium strain ZB-16A was screened out from them.Studies of normal morphology,physiological and biochemical characterizing,16S rDNA gene sequence and phylogenesis tree analyses indicated that strain ZB-16A belongs in Pseudomonas putida.ZB-16A tolerates 6 000 mg/L nicotine concentration and completely degrades nicotine in 48 hours at 30℃ when initial pH of culture medium and rotation speed of shaker are 7.0 and 150 rpm,respectively;while the growth of ZB-16A is inhibited at an initial nicotine concentration of 7000 mg/L.
