植物源抗烟草花叶病毒活性物质的初步筛选

Preliminary Screening of Bioactive Substances from Plants Against Tobacco Mosaic Virus

  • 摘要: 利用半叶枯斑法,测定了13科18种植物蛋白粗提液对烟草花叶病(TMV)的抑制效果。结果表明,不同植物蛋白粗提液对TMV侵染心叶烟的抑制效果存在差异,6种植物蛋白粗提液对TMV的抑制效果达到50%以上,其中菠菜和合欢最高,分别达到了97.44%和96.92%。进一步通过(NH4)2SO4盐析,对菠菜和合欢蛋白粗提液抗病毒活性成分进行了初步分离,并测试(NH4)2SO4盐析的不同蛋白组分对TMV的抑制效果。结果显示,菠菜蛋白粗提液盐析后的两个蛋白组分都表现出很高的抗病毒活性,而合欢蛋白粗提液只在40%~80%饱和度盐析的蛋白组分具有较强的抗病毒活性。

     

    Abstract: The antiviral activities against TMV of crude protein extracts from 18 plants of 13 families were determined by local lesion assay.The results showed that the inhibition effects of different protein extracts on TMV infected Nicotiana glutinosa were varied.The inhibition rates of six extracts were over 50%,among them,those of Spinacia oleracea and Albizzia julibrissin Durazz extracts reached 97.44% and 96.92%,respectively.The crude extracts from Spinacia oleracea and Albizzia julibrissin Durazz were further isolated primarily by ammonium sulfate precipitation,and the inhibition effects of the resulted protein fractions were determined.The results showed that the two protein fractions from Spinacia oleracea extract presented very high antiviral activity;for Albizzia julibrissin Durazz extract,only the fractions precipitated from ammonium sulfate at 40%~80% saturation showed relatively high antiviral activity.
