烟梗废料液态发酵生产苏云金芽孢杆菌的适宜条件筛选

Producing Bacillus thuringiensis by Liquid State Fermentation of Waste Stem

  • 摘要: 以烟草废料烟梗为原料,采用正交组合设计方法,对苏云金杆菌HD-1菌株(Bacillus thuringiensis HD-1)的液态发酵培养基配方和发酵适宜条件进行摇瓶优化筛选。结果表明,烟梗液态发酵培养基的最佳组合为:烟梗液态基础培养基添加1.5%的蛋白胨,0.4%的酵母膏,0.5%的葡萄糖,0.1%的磷酸二氢钠。发酵适宜条件为:接种量在1%,pH 7,温度30℃,摇床转速180 r/min。烟梗发酵生产苏云金芽孢杆菌杀虫剂的方法是可行的,为烟草废料的循环利用开辟了新途径。

     

    Abstract: Using waste stems as material,the liquid culture medium formula of Bacillus thuringiensis HD-1 and fermentation conditions were optimized by shaking flask culture with orthogonal experiments.The results showed that the optimum culture medium formula was:basic liquid tobacco stem culture medium adding peptone 1.5%,yeast extract 0.4%,glucose 0.5%,and di-sodiumhy drogerphosphate 0.1%.The preferable condition for fermentation was:volume of inoculation 1%,pH 7.0,temperature 30℃ and shaker speed 180 r/min.The production method of Bacillus thuringiensis insecticide by tobacco stem fermentation is feasible,it opened up a new usage for tobacco waste.
