  • 摘要: 为进一步了解和认识CuZnSOD基因的结构和功能,从13个烟草品种及林烟草种的幼叶cDNA中成功扩增出CuZnSOD基因编码区全长序列,并对其进行序列分析。烟草CuZnSOD基因编码序列全长291bp,编码1个96个氨基酸长度的蛋白质,其核苷酸序列关键区域与Genbank中其他植物的CuZnSOD相似性高至94%。遗传进化分析结果表明,烟草CuZnSOD基因与西红柿和马铃薯CuZnSOD基因亲缘关系最近。通过荧光定量PCR对低温弱光胁迫条件下13个烟草品种及林烟草种的CuZnSOD基因表达量进行分析,结果显示在低温弱光胁迫条件下,CuZnSOD基因在13个烟草品种及林烟草种间表达模式存在显著差异。对烟草CuZnSOD基因表达模式的分析,有助于揭示烟草对环境的适应机制及抗氧化的生理机制。

     

    Abstract: For further understanding the structure and function of CuZnSOD gene in Nicotiana tabacum, the full-length sequence of CuZnSOD gene encoding zone was successfully amplified from young leaves of 13 tobacco cultivars and Nicotiana sylvestris, and its sequence was analyzed. It was revealed that the CDS contained 291 base pairs and encoded a protein of 96 amino acids. The results of key nucleotide sequence comparison showed that the similarity of CuZnSOD genes with other plant species in Genbank was up to 94%. Meanwhile, the analysis of phylogenic tree based on CuZnSOD CDS of tobacco indicated its closest genetic relationship with tomato and potato. The results of quantitative PCR analysis of the expression levels of CuZnSOD showed significant differences in expression pattern between low-temperature and weak-light exposure samples of 13 cultivars and Nicotiana sylvestris. The expression pattern analysis of CuZnSOD gene in tobacco would provide clues to the revelation of the adaptation mechanism of tobacco to environment, as well as the physiological mechanism of anti-oxidation.
