烟草中蛋白质含量的快速测定

Rapid Determination of Protein Content in Tobacco

  • 摘要: 为了快速测定烟草中的蛋白质含量,利用乙酸加热提取的方式,使非蛋白质含氮组分与水溶性蛋白质分离,再采用连续流动分析仪,以紫外光催化、在线加压升温消化的方式,测定烟草中的非蛋白质氮含量,进而计算求得烟草的蛋白质含量。结果表明:①该方法检测结果稳定,变异系数小于3%;②其测定值与现有标准测定结果无显著差异,T检验结果为t=1.082,小于t0.1,15=1.753;③该检测方法可靠,对非蛋白质氮的检测回收率在95%~101%范围内。

     

    Abstract: In order to rapidly determine the protein content in tobacco,the compositions of non-protein nitrogen was separated from water-soluble proteins by heating with acetic acid,and the content of non-protein nitrogen in tobacco was determined with continuous flow analyzer combining with UV catalytic device and online pressurized heater,then the content of protein in tobacco was calculated on the basis of measured non-protein nitrogen content.The results showed that:1) The detection results were stable with CV less than 3%;2) There was no significant difference between the detection results of this method and the current standard method,the result of T test was t=1.082,which was less than t0.1,15=1.753;3) This method was reliable with the determining recoveries of non-protein nitrogen from 95% to 101%.
