Effects of Extract Solution and Standard Solution on Determination of Contents of Glucose,Fructose,Sucrose and Maltose in Tobacco

  • 摘要: 为探究萃取液和标准制备液对烟草中葡萄糖、果糖、蔗糖和麦芽糖4种糖含量测定的影响,比较了水和氢氧化钠水溶液作溶剂萃取烟草样品中4种糖的稳定性,以及所配制标准制备液的稳定性。结果表明:①0.02 mol/L氢氧化钠溶液萃取烟草样品,可有效抑制提取液中蔗糖和麦芽糖的水解,葡萄糖、果糖、蔗糖和麦芽糖在16 h内的含量与初始值的偏差小于15%;②用水配制标准制备液,30 d内储备液的相对标准偏差在1%以内,能够保证定量准确。用氢氧化钠水溶液萃取烟草样品和用水配制标准制备液有助于准确测定烟草中葡萄糖、果糖、蔗糖和麦芽糖的含量。

     

    Abstract: To investigate the effects of extract solution and standard solution on the determination of contents of glucose,fructose,sucrose and maltose in tobacco samples,the stabilities of the four sugars in the extract solutions of water or aqueous sodium hydroxide were compared,and the stability of prepared standard solution was also determined.The results showed that:1) The hydrolysis of sucrose and maltose in extract solutions was effectively inhibited while 0.02 mol/L aqueous sodium hydroxide was used for extracting tobacco samples.The variations of the contents of the four sugars were less than 15% within 16 hours.2) The quantitative accuracy could be ensured with the relative standard deviation of stock solution less than 1% within 30 days when water was used for preparing the standard solution.The contents of glucose,fructose,sucrose and maltose in tobacco could be accurately determined by using aqueous sodium hydroxide solution as the extract solvent and using water for preparing the standard solution.
