烟草黄瓜花叶病毒湖北分离物全基因组序列分析及亚组鉴定

Complete Sequence Analysis and Subgroup Identification of Cucumber mosaic virus from Nicotiana tabacum in Hubei

  • 摘要: 为明确我国湖北烟区烟草黄瓜花叶病毒(Cucumber mosaic virus,CMV)的亚组类型,对湖北地区侵染烟草的CMV HB24株系进行基因组全长克隆和序列分析。结果表明,HB24株系RNA1(GenBank序列号:KC019299)全长为3363个核苷酸(nt),编码993个氨基酸的1a蛋白;RNA2(GenBank序列号:KC019300)全长为3049nt,编码859个氨基酸的2a蛋白和111个氨基酸的2b蛋白;RNA3(GenBank序列号:KC019301)全长为2216nt,编码279个氨基酸的3a蛋白和218个氨基酸的CP蛋白。核苷酸序列同源性分析表明,HB24与Fny,SD和Q株系RNA1同源性分别为91.6%,91.1%和75.9%,RNA2同源性分别为91.8%,97.8%和69.5%,RNA3同源性分别为92.1%,93.4%和73.2%;与同属花生矮化病毒(Peanut stunt virus,PSV)ER株系RNA1,RNA2和RNA3的同源性分别为65.5%,56.7%和54.1%,与同属番茄不孕病毒(Tomato aspermy virus,TAV)V株系同源性分别为66.9%,57.7%和51.5%。HB24 RNA35'NTR(Nontranslated region)的结构分析以及RNA35'NTR和CP基因核苷酸序列系统进化树的分析表明,HB24属于CMV亚组IB。

     

    Abstract: In order to identify the subgroup type of CMV(Cucumber mosaic virus) in Hubei tobacco,CMV HB24 strain was isolated,and the full-length cDNA sequence of genomic RNAs of the isolate was determined by RT-PCR and sequence analysis.The results showed that:HB24 RNA1(GenBank:KC019299) consisted of 3363 nucleotides(nt) and encoded 1a protein of 993 amino acid residues(aa),RNA2(GenBank:KC019300) consisted of 3049 nt and encoded 2a protein of 859 aa and 2b protein of 111 aa,RNA3(GenBank:KC019301) contained 2216 nt and encoded 3a protein of 279 aa and CP of 218 aa.The results of nucleotide sequence similarity indicated that:the similarities of HB24 RNAs with the RNAs of Fny,SD,Q strains were:RNA1:91.6%,91.1%,75.9%;RNA2:91.8%,97.8%,69.5%;RNA3:92.1%,93.4%,73.2%,respectively.The similarities of HB24 RNAs with the RNAs of PSV-ER(Peanut stunt virus) were RNA1:65.5%,RNA2:56.7%,RNA3:54.1%;and that with the RNAs of TAV-V(Tomato aspermy virus) were 66.9%,57.7%,51.5%,respectively.According to RNA3 5' NTR(Nontranslated region) structure analysis,and phylogenetic tree analyses of RNA3 5'NTR and CP gene nucleotide sequences,HB24 belonged in CMV IB subgroup.
