四川省烟草白绢病病原菌的分离鉴定及其生物学特性

Identification of Pathogen of Tobacco Southern Blight(Sclerotium rolfsii Sacc.) in Sichuan and its Biological Characteristics

  • 摘要: 烟草白绢病是近年来在四川烟区发现的新病害,为明确其病原特征,进而有效防治该病害,在烟田采集了烟株根部菌丝,进行分离、纯化,并命名为B1。并进行了病原菌形态观察,18S rRNA鉴定及致病性测定。结果表明,B1为半知菌亚门,丝孢纲,无孢目,小菌核属的齐整小核菌Sclerotium rolfsii Sacc。生物学特性研究表明,白绢病菌B1菌丝和菌核致死温度为50℃处理10 min,且菌丝更适合在偏酸性条件下生长,最适pH为5.0。

     

    Abstract: Southern blight was a new tobacco disease found in Sichuan,to clarify its pathogenic characteristics and control it effectively,the hypha of the pathogen was collected from tobacco roots in field,it was isolated, purified and named as B1.By means of morphological observation,18S rRNA sequence analysis and pathogenic test,B1 was identified as Sclerotium rolfsii Sacc,which belongs in subphylum Deuteromycotina,class Hyphomycetes,order Agonomycetales,genus Sclerotium.The results of biological characteristics test showed that the lethal temperature of hypha and sclerotium of B1 was 50℃ for 10 minutes,acidic condition was preferable to hypha growth,the best was pH 5.0.
