Determination of Contents of Amylose and Amylopectin in Tobacco by Dual Wavelength Spectrophotometry

  • 摘要: 根据双波长比色原理,选择直链淀粉的测定波长为512和596 nm,支链淀粉的测定波长为560和695 nm,依据回归方程求出直链和支链淀粉的含量。结果表明:用双波长分光光度法测定直链淀粉和支链淀粉的含量结果准确,适用于批量分析。

     

    Abstract: Determining the contents of amylose and amylopectin in tobacco with dual-wavelength spectrophotometry was new, although it has been widely applied to other crops. The measurement wavelengths of 512 and 596 nm for amylose and 560 and 695 nm for amylopectin were selected according to the principle of dual wavelength colorimetric, and the contents of amylose and amylopectin were calculated based on regression equations. The results showed that the contents of amylose and amylopectin in tobacco determined with dual-wavelength spectrophotometry were accurate, and the method was suitable for the determination of amylose and amylopectin in batches of tobacco samples.
