微生物制剂MP喷施烟叶后的差异蛋白分析

Analysis of Differential Protein in Tobacco Leaves Sprayed with Microbial Agent MP

  • 摘要: 为明确微生物制剂MP(芽孢杆菌)喷施烟叶后对其品质的影响,以喷施微生物制剂MP和清水的烟叶作为研究材料,采用蛋白质双向凝胶电泳法分析了烟叶中蛋白质的表达差异。结果显示,在MP制剂诱导烟叶后,有6个蛋白质的表达量发生了显著变化,其中5个蛋白点上调表达,1个蛋白点下调表达。利用基质辅助激光解析串联飞行时间质谱法(MALDI TOF/TOF MS)对这6个差异蛋白进行了分析,结果共鉴定出4个差异蛋白,分别为烟草酸性几丁质酶PR-P、烟草几丁质酶前体、内切几丁质酶A前体和逆渗透蛋白。几丁质酶和逆渗透蛋白与植物的抗病性密切相关,说明在喷施MP制剂后烟叶可能产生抵御外源病菌的应急机制并进而提高其抗病性。

     

    Abstract: In order to find out the effects of Microbial agent MP(Bacillus magaterium)spraying on the quality of tobacco leaves, two-dimensional gel electrophoresis(2-DE)was used to analyze the differences of protein expression between tobacco leaves sprayed with MP and water respectively. The results revealed that the expression levels of six proteins changed significantly, among them the expression levels of five protein points increased, and that of the other one decreased after MP treatment. The six differential proteins were identified with matrix-assisted laser desorption ionization time of flight mass spectrometry(MALDI TOF/TOF MS), the results showed that four of them were recognized as tobacco acidic chitinase PR-P, tobacco chitinase precursor, endochitinase A precursor and osmotin respectively. Chitinase and osmotin closely related to plant's disease resistance, it indicated that the emergency system against exogenous bacteria might be induced and by which disease resistance was improved after MP spraying.
