Determination of Pectin in Tobacco with 3, 5 -Dinitrosalicylic Acid Colorimetry

In order to determine the pectin in tobacco accurately, the sample was selectively degraded by pectinase, and determined with 3, 5-dinitrosalicylic acid colorimetry.The result of this method was compared with that of using carbazole as a chromophoric reagent.The results showed that the recovery of pectin was 96.30%-100.20%with the RSD of 0.98%-1.85% in the linear range of 50-1500μg/ml and R²=0.9991.The content of pectin in tobacco determined by this method was comparable with that determined by the carbazole method.
