Using flue-cured tobacco cv.NC2326, G28 and their hybrid F1 as materials, the effects of major elements in PCR reaction system on SSR analysis were studied.The results showed that under the conditions of 1.5U TaqDNA polymerase, terminal MgCl2 concentration of 1.5mmol·L-1, 200μmol·L-1 dNTPs, 0.25μmol·L-1 primers and 20ng templates of DNA in total volume of 20μL, the result was better in PCR reaction.
