Determination of Pectin in Tobacco with Enzymolysis-Autoanalyzer

A method for determining the pectin in tobacco with enzymolysis-autoanalyzer was developed, and the enzymolysis conditions were optimized.The tobacco powder was extracted with 80% alcohol at reflux for 1hr, then filtrated.The resultant residue was enzymolyzed with 480u/g of pectinase at 58℃ for 2hrs in 60mL of sodium citrate/citric acid (pH4)buffer solution.The galacturonic acid from pectin degraded in tobacco was determined with an autoanalyzer.The method has good linearity with a correlation coefficient of 0.9997.The recovery of pectin in tobacco was from 96.8% to 101.0%, and the RSD of the method was 1.70%.
