The DNA Fingerprints and Varieties Identification of Tobacco Seed

Abstract
Random amplified polymorphic DNA (RAPD)is a popular technique of fingerprinting.Good quality DNAs were extracted from 10 varieties accessions of tobacco by using improved "ROSE" method.Reproducible amplification products were obtained with rigorously optimized reaction conditions.Fifty-three amplification fragments were generated by RAPD with 10 primers, among which 10 fragments were shared by all 10 accessions.The degree of polymorphism was 81.1%. RAPD patterns of 10 accessions generated by P7 could be used as DNA fingerprint for identification of 10 tobacco varieties.
